Serodiagnosis of Leptospirosis by Enzyme-linked Immunosorbent Assay and Flow Cytometric Analysis

Abstract
ELSA & Flow cytometric analysis were used to serodiagnosis of leptospirosis. Leptospira interrogans serovar mwogolo were evaluated the sensitivity and specificity of the analysis. The reactivity of IgG and IgM IgM antibodies in the sera from
patients of
leptospirosis and normal healthy control were analysed with the phenol extracted cell antigen. The reactivity(absorbance at 409 nm) of IgG and IgM in the sera of patients with leptospirosis to L. interrogans serovar mwogolo antigen was

and
1.208¡¾0.072 respectively. Correlation coefficient between the MAT and ELIST O. D. of IgM to the antigen was 1.22 Absorbance above 0.15 was determined positive for the diagnosis of leptospirosis, the sensitivity and specificity of IgG was 100%
and
98%,
respectively. The optical density of the positive sera ELISA was 1.79 in IgG and 1.84 in IgM at 1:100 serum dilution and was 0.15 in IgG and 0.12 in IgM at 1:6400 serum dilution. But, histogram with the peak modal channel of Flow cytometric
analysis not
varied greatly.
